Insufficient activity or stability of exogenous enzymes in chassis cells is one of the bottlenecks in the biosynthesis of natural products. Through random mutation and enzyme-based protein structure design of targeted mutations for substrate binding sites or active centers, the catalysis of key enzymes can be improved. Activity, thermal stability, pH tolerance, solubility, and tolerance to salts and organic solvents improve yields of target products.
